ThrombocytinThrombocytin is a single-chain glycopeptide from Bothrops atrox venom. It splits several synthetic chromogenic substrates, such as Tos-Gly-Pro-Arg-pNA or Bz-Phe-Val-ArgpNA, and its action is inhibited by diisopropyl fluorophosphate (DFP), soybean trypsin inhibitor and antithrombin III-heparin complex. Thrombocytin does also activate factor V by limited proteolysis. Thrombocytin induces platelet aggregation in platelet-rich plasma and activates the contractile system of platelets. Thrombocytin-induced platelet aggregation is inhibited by prostaglandin E, antithrombin III and heparin, however, not by the specific thrombin inhibitor hirudin. Thrombocytin may therefore be used for testing platelet function in plasma of hirudintreated patients and laboratory animals. |
