Platelets Lyoph.Lyophilized platelets are a preparation of fixed human platelets which have been lyophilized for long-term stability. Each vial of lyophilized platelets is reconstituted with the appropriate volume of Tris Buffered Saline to yield a platelet count of approximately 275,000 per ul. The Ristocetin Cofactor Activity in human plasma may be determined by the agglutination of a standardized platelet suspension in the presence of vonWillebrand factor using the antibiotic, Ristocetin. Although the platelets play a passive role in the agglutination, there is an absolute requirement that the Ristocetin-dependent receptor to which vonWillebrand factor binds be intact. It is generally accepted that the Ristocetin Cofactor Activity is the most useful in vitro assay for the diagnosis of vonWillebrands disease. |
